Objective: To study the role of palmitoleic acid (PA) in the pathogenesis of acute anterior uveitis (AAU). Methods: The levels of PA in the stools of patients AAU measured by gas chromatography coupled with mass spectrometry (GC-MS) and compared with samples obtained from healthy individuals. Enzyme Linked Immunosorbent Assay (ELISA) and flow cytometry (FCM) was used to assess the effects of PA on dendritic cells (DC) and CD4 + T cells obtained from mice, AAU patients and healthy individuals. C57BL / 6 mice were fed with PA or vehicle and experimental autoimmune uveitis (EAU) was induced by human retina IRBP651-670 peptides.
Disease severity was evaluated with clinical manifestations of EAU and histology. Spleen cell differentiation of type 1 T helper (Th1) and Th17 cells was evaluated by FCM. tandem mass tags (TMT) based proteomics analysis was used to identify proteins expressed differently following the incubation of DC with the PA.
Results: The concentration of droppings from AAU PA increased in patients compared with healthy people. In vitro, PA promoted apoptosis of DC and inhibit the secretion of TNF-α from mouse cells bone marrow-derived dendritic (BMDCs) as well as DC from AAU patients and healthy individuals. Just downhill DC surface marker expression and IL-12p70 secretion in BMDCs and healthy individuals but not in DC DC AAU patients. PA-treated BMDCs inhibited Th cell differentiation from naive mice cells CD4 + T cells and IL-17 and IFN-γ secretion in co-culture supernatant. PA also inhibit Th cell differentiation and secretion of IFN-γ and IL-17 in CD4 + T cells from mice, AAU patients and healthy individuals. In vivo, PA-treated EAU mice showed clinical and histopathological manifestations of intraocular lighter than the control group.
PA eat spleen inhibits Th17 cell differentiation, whereas Th1 cells are not affected. Up to 30 upregulated and downregulated 77 proteins were identified when comparing the PA-treated DC with control. Conclusions: An increased expression of PA feces observed in patients AAU. PA proved to have immunoregulatory effects on DC cells and CD4 + T and attenuated the severity of the disease in EAU mice.

chronic stress and accelerated aging has been shown to have an impact on the inflammatory response and related outcomes such as sepsis and organ failure, but data is lacking in the literature of trauma. The purpose of this study was to investigate the potential link between stress and outcome pretrauma posttrauma. the hypothesis is that chronic stress pretrauma accelerate aging, which increases susceptibility to posttrauma sepsis and organ failure. In the prospective, this correlational study, chronic stress and premature aging biological compared with systemic inflammatory response syndrome, sepsis and organ failure in trauma patients aged 18-44 years.
The results support the hypothesis with a significant overall association between susceptibility to sepsis and premature biological aging (n = 142). There was also a significant negative association between cytokine levels mean and chronic stress. The strongest association was found between average interleukin-1β ( IL -1β) and Human telomerase reverse transcriptase (hTERT), r (101) = -0.28), p = , 004. significant negative association was found between the levels of cytokines mean, IL – 12p70 , r (108) = -0.20, p = 0.034; and tumor necrosis factor-α (TNF-α), r (108) = -0.20, p = 0.033, and positive life events through behavioral measure chronic stress.
