In addition to helper T cells (Th), natural killer cells (NK) have also been explained to participate in the formation of Dendritic Cell (DC) cells – adaptive immune response. At present, it is still unclear to the extent to which the induction of the Helper NK cell immune mechanism is coupled with th response and whether the two immune response helper caused by the same DC on the same receptor receptor stimulation (PRR) receptor. In this study, we showed that the maturation of DC with a cocktail containing the FMKP (membrane fragments of Klebsiella pneumoniae) increased both cell cells and NK helper cells in the way that triggered the PRR dose as determined by the capacity of the helper until it produces IFN-γ.
Furthermore, by triggering additional PRR paths [FMKP in combination with poly (I: c) Lyovec], we reveal that both approaches modulate the number of IL-12P70 DC-12P70 and that this cytokine is the main determinant of DC-induced of the DC cell helper response TH1 and NK. In addition, all PRR triggers are able to induce IL-12-producing adult DC enough to encourage this helper response. We propose the existence of one program used by DC to induce strong cellular immune responses by stimulating the process of T helper and NK. This knowledge can help choose the right PRR trigger in preventive and therapeutic vaccine design.
Activation and profile of the cytokines of dendritic cells that are lowered in leprosy: in vitro stimulation by Sonicated Mycobacterium Leprae induce a decrease in the level of <em> il </ em> 12p70 </ em> on lepromatous leprosy.
Dendritic cells (DCS) play an important role in the default relationship and host’s adaptive immunity to mikobacterial infections. The study of the interaction of DC monocyte-derivative (MO-DCS) using Mycobacterium leprae in leprosy patients rarely occur. This study shows that MOS differentiation to similar DCS in all leprosy forms is compared to healthy individuals. In vitro stimulation from MO-DC which is immature with Sonicated M. Leprae induces the degree of DC maturation variables as determined by the increase in HLA-DR, CD40, CD80 and CD86 expressions, but not CD83, in all groups studied. Different cytokine production by MO-DC appears similar in all groups studied in similar conditions.
However, the production of Interleukin (IL) -12P70 by MO-DCS from Lepromatous leper patients (LL) after in vitro stimulation with M. Leprae is lower than tuberculoid leper patients and healthy individuals, even after CD40 ligation with CD40 ligand transfection cells , Cumulative findings today show that MO-DCS from LL patients is generally an IL-12P70 producer which is weak despite having the activation properties of moderate OFM. Leprae. These results can explain the immunity M. Leprae-specific is poor in the type of leprosy LL.
Mycobacterium tuberculosis promotes TH17 expansion through human dendritic cell regulations towards CD14 and Low <em> il </ em> 12p70 </ em> phenotypes that are reprogrammed with IFN-γ.
The capacity to develop protective immunity to heterogeneous mikobakteria is distributed among humans, and is currently unknown why the initial immune response induced by Mycobacterium tuberculosis (MTB) does not provide the right distance from this pathogen. Dendritic cells (DCS) are some of the first cells that interact with MTB and they play an important role in developing protection immunity to MTB. Given that MTB’s infected macrophages have difficulty condescending MTB, they need help from CD4 + IFN-γ cells that are reproduced through DC which produces IL-12P70.
Here we report that MTB modified the plasticity of human DC by expanding a subset of DC CD14 + with a weak IL-12P70 production capacity. Subset promoted MTB CD14 + further from IFN-γ with CD4 + naive T cells, but instead pushes RORT + CD4 + CD4 + IL-17A cells. Peptidoglycan which is revealed MTB and Lipoarabinomanan Mannosylated some recapitulate subset partitions caused by MTB. The addition of IFN-γ, but also IL-17A or IL-22, which has the potential to be produced by cells γ / δ-T which is exposed to MTB in the mucous layer, inhibiting differentiation on CD14 + DCS and promoting high-level IL-12P70 in MTB – Help DC. We conclude that MTB exploits DC plasticity to reduce IL-12P70 production, and that this process can be fully ignored by IFN-γ exogenous. These data show that strategies for increasing the production of local IFN-γ in the lungs of tuberculosis patients can increase host immunity to MTB.

Monitoring TNFR1, <em> il </ em> -2rα, HGF, CCL8, <em> il </ em> -8 and <em> il </ em> – <em> 12p70 </ em> following HSCT and role They are GVHD biomarkers in pediatric patients.
There are no predictive factors currently available to set the risk of the patient’s special GVHD. Six Serum Cytokine Panel (TNF 1 Receptor, Alfa Receptor IL-2 (IL-2Rα), Hepatocyte Growth Factor (HGF), Protein-2 Monocyte Chemo-2, IL-8, IL-12P70) is monitored at the time set by points ( Day -1, +1, +7, +14, +21, +28 and +60) In 170 SCT Hematopoietic Pediatric Recipients (HSCT). We found that higher IL-2Rα concentrations on day +14 and +21 together with HGF on +14 and +21 were significantly related to the higher probability of the second class II-IV GVHD (on the day +14 : 60% vs 28%, p = 0.007) and class III-IV (on the day +14 it is: 40% vs 15%, p = 0.001).

Description: Description of target: The protein encoded by this gene is a secreted cytokine that is important for the proliferation of T and B lymphocytes. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli.;Species reactivity: Human;Application: ;Assay info: Assay Methodology: Quantitative Sandwich ELISpot Immunoassay;Sensitivity: |

Description: Description of target: The protein encoded by this gene is a pleiotropic cytokine produced by activated T cells. This cytokine is a ligand for interleukin 4 receptor. The interleukin 4 receptor also binds to IL13, which may contribute to many overlapping functions of this cytokine and IL13. STAT6, a signal transducer and activator of transcription, has been shown to play a central role in mediating the immune regulatory signal of this cytokine. This gene, IL3, IL5, IL13, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL13. This gene, IL13 and IL5 are found to be regulated coordinately by several long-range regulatory elements in an over 120 kilobase range on the chromosome. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported.;Species reactivity: Human;Application: ;Assay info: Assay Methodology: Quantitative Sandwich ELISpot Immunoassay;Sensitivity: |

Description: Description of target: This gene encodes a cytokine that functions in inflammation and the maturation of B cells. In addition, the encoded protein has been shown to be an endogenous pyrogen capable of inducing fever in people with autoimmune diseases or infections. The protein is primarily produced at sites of acute and chronic inflammation, where it is secreted into the serum and induces a transcriptional inflammatory response through interleukin 6 receptor, alpha. The functioning of this gene is implicated in a wide variety of inflammation-associated disease states, including suspectibility to diabetes mellitus and systemic juvenile rheumatoid arthritis. Alternative splicing results in multiple transcript variants.;Species reactivity: Human;Application: ;Assay info: Assay Methodology: Quantitative Sandwich ELISpot Immunoassay;Sensitivity: |

Description: Description of target: This gene encodes a cytokine that acts as a growth and differentiation factor for both B cells and eosinophils. The encoded cytokine plays a major role in the regulation of eosinophil formation, maturation, recruitment and survival. The increased production of this cytokine may be related to pathogenesis of eosinophil-dependent inflammatory diseases. This cytokine functions by binding to its receptor, which is a heterodimer, whose beta subunit is shared with the receptors for interleukine 3 (IL3) and colony stimulating factor 2 (CSF2/GM-CSF). This gene is located on chromosome 5 within a cytokine gene cluster which includes interleukin 4 (IL4), interleukin 13 (IL13), and CSF2 . This gene, IL4, and IL13 may be regulated coordinately by long-range regulatory elements spread over 120 kilobases on chromosome 5q31.;Species reactivity: Human;Application: ;Assay info: Assay Methodology: Quantitative Sandwich ELISpot Immunoassay;Sensitivity: |

Description: Description of target: The protein encoded by this gene is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Mutations in this gene are associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis.;Species reactivity: Human;Application: ;Assay info: Assay Methodology: Quantitative Sandwich ELISpot Immunoassay;Sensitivity: |

Description: Description of target: This gene encodes a subunit of a cytokine that acts on T and natural killer cells, and has a broad array of biological activities. The cytokine is a disulfide-linked heterodimer composed of the 35-kD subunit encoded by this gene, and a 40-kD subunit that is a member of the cytokine receptor family. This cytokine is required for the T-cell-independent induction of interferon (IFN)-gamma, and is important for the differentiation of both Th1 and Th2 cells. The responses of lymphocytes to this cytokine are mediated by the activator of transcription protein STAT4. Nitric oxide synthase 2A (NOS2A/NOS2) is found to be required for the signaling process of this cytokine in innate immunity.;Species reactivity: Human;Application: ;Assay info: Assay Methodology: Quantitative Sandwich ELISpot Immunoassay;Sensitivity: |

Higher Serum IL-8 concentration on the day +28 is associated with a lower probability of chronic GVHD to 4% vs 29% (p = 0.01) for patients with lower serum IL-8 concentrations vs. This finding is confirmed when the analysis is limited to the non-related donor group. In conclusion, even if serum cytokine levels are associated with several variables related to HSCT, we identify two cytokines as predictors of GVHD II-IV and III-IV, translate into higher TRM risks (17% vs 3%, p = 0.004 ).
