Recently there was a paradigm shift in the field of Dendritic immunotherapy (DC) Dendritic, where several clinical studies have confirmed feasibility and excellence using DCS originating from human blood isolated directly above distinguished vitro. There are two main DC sets found in the blood; Plasmacytoid DCS (PDC) and Myeloid DCS (MDCs), and both have been clinically tested. CD1C + MDC is a cell that presents a highly efficient antigen that has the ability to issue IL-12P70, while PDC is the cells that are excluded professionally ifn-α which is proven to induce default immune response in melanoma patients.
Therefore, combining MDC and PDCs poses as an attractive multi-function vaccine approach. However, the type of IFNS has been reported to inhibit the production of IL-12P70 and MDC-induced MDC cell activation. In this study, we investigate the effect of IFN-α in the maturation and MDC function. We show that the two IFN-α PDC is recuminated and activated greatly increases MDC maturation and increases IL-12P70 production. MDC and PDCs that are cultured together also have beneficial effects on NK activation and cells and also increases IFN-γ production with allogeneic t cells. Conversely, the existence of type I IFN reduces proliferative cell response. The presence is only a small part of PDC active enough for this effect and the ratio needed between the set of insecure parts. Taken together, these results support the use of MDC and PDC combined into one immunotherapy vaccine with a broad immunostimulator feature.

Vector bacteria has been proposed as a new vaccine strategy to encourage strong cellular immunity. Entitulated Brucella strains consist of a promising vector candidate because they have the potential to induce the response of immune mediated CD4 (+) and CD8 (+) which are strong with the absence of excessive inflammation such as observed with other gram-negative bacteria. However, some strains of Brucella disturb the maturation of dendritic cells (DCS), which are important for the priming of antigen specific T cells. In this study, we investigated DC interactions which were derived from the human monocyte smoothly weakening B. Abotor strain 19, which had previously been employed successfully to vaccinate livestock.

We first saw the potential of S19 to inhibit the maturation of DC caused by cytokines; However, infected cells are expressed by CD25, CD40, CD80, and CD86 to the extent that are comparable as DC that is not infected and mature. Furthermore, S19 activates DCS in the absence of exogenous stimuli, increases HLA-ABC and HLA-DR expression, and is able to survive intracellularly without causing cytotoxicity. Thus, DCS provides a cellular niche for Brucella which survives in vivo as a permanent source of antigen. S19-infected DCS produces IL-12 / 23P40, IL-12P70, and IL-10, but not IL-23. While hot bacteria also activate DCS, the soluble mediator is not involved in the activation of S19 induced human DC. HEK 293 Transfectants revealed cellular activation by S19 mainly through receptor involvement such as toll (TLR) 2. Thus, as an immunological prerequisite for the efficacy of vaccines, B. S19 abortion has the potential to infect and potentially activate (most likely through TLR2) human DCS to produce cytokines that promote TH1.

A previous study revealed the relationship between the vulnerability to Takayasu arteritis (no) and one nucleotide polymorphism (SNP) RS6871626 located in IL12B, which encodes Interleukin (IL) -12P40, general components of IL-12P70 and IL-23. We investigate this cytokine expression in patients with no, emphasizing them into them with or without risk of alleles at SNP RS6871626.
The Level of Plasma IL-12P40, IL-12P70, and IL-23 were quantified in 44 patients with no and 19 healthy controls (HCS) by testing enzyme immunosorbents. Monocytes are obtained from 20 patients with no and 14 HCS, treated with interferon-γ (IFN-γ) and lipopolysaccharide, and then the cytokine supernatis is quantified. In addition, IFN-γ + or IL-17A + cell ratios to CD4 + cells are measured by the analysis of cytometric flow of peripheral mononuclear blood cells.
